چکیده

A simple radioligand assay has been developed for measuring cortisol in plasma and urine. The antiserum was produced in rabbits by using cortisoi-21hemisuccinyl-bovine serum albumin as the antigen. The standard curve was linear from 0-1000 pg. A small aliquot of ethanol-precipitated plasma or urine is directly assayed. Interassay variability of 56 separate assays of a normal plasma pool during six months was: mean, 224 zg/iiter; range, 210-238 pg/liter; CV, 3.2%. In assay of multiple samples obtained between 0800 and 1800 hours from normal young men, radioimmunoassay (AlA) gave consistently lower values than did competitive protein binding (CPB). The relationship, as calculated from resuits for 41 single samples, is expressed by the equation RIA = 0.713 CPB + 0.59, suggesting that the normal range for RIA should be 35-180 pg/liter rather than the 50-250 ag/liter usually cited. Although there is crossreactivity with li-deoxycortisol, this assay is clearly superior to CPB: it is more specific, has a wider range (<8-400 pg/liter), and requires less technician time.

Rapid, specific assay for plasma cortisol by competitive protein binding.

We describe a modified competitive protein-binding method for assay of plasma cortisol. Plasma samples are deproteinized by dilution with an ethanol/phosphate buffer, followed by heating at 100 degrees C for 2 min. Horse serum is used as the source of transcortin. Free radioactivity is separated from the protein-bound component by partition into liquid scintillation counting within 60 min. The ...

Comparison of solid-phase radioimmunoassay and competitive protein binding method for postdexamethasone cortisol levels in psychiatric patients.

Results obtained by competitive protein binding assay (PBA) and a solid-phase radioimmunoassay (RIA) for cortisol were compared in 157 samples from 100 psychiatric patients given a dexamethasone suppression test (DST). Cortisol levels in plasma samples obtained at 8:00 a.m. or 4:00 p.m. the day following 1.0 mg dexamethasone orally at bedtime ranged from 0 to 30 micrograms/dl and correlated clo...

Comparison of clinical assays for serum corticosteroids.

We compared results obtained by (a) chemical (spectrophotometric) and competitive protein-binding assays for serum corticosteroids, and (b) competitive protein-binding and radioimmunoassay, with and without CCI-4 extraction, for serum cortisol. The two corticosteroid methods gave identical results. Radioimmunoassay with and without a CCI-4 extraction step gave identical results for cortisol. Co...

Parallel radioimmunoassay for plasma cortisol and 11-deoxycortisol.

We describe a direct, rapid, and specific procedure for the parallel radioimmunoassay for cortisol and 11-deoxycortisol in plasma. The plasma sample is used directly, after heat inactivation of the natural cortisol-binding protein. The radioimmunoassay utilizes antibodies generated in rabbits by steroids congugated at their 3-oxo position to thyroglobulin. Ammonium sulfate is used to separate b...

Determination of adrenal response after oral administration of multiple doses of methylprednisolone.

ORMAL cortisol production by the adrenals has a circadian rhythm, and many studies1-6 have shown a variation of plasma cortisol with the time of day. Migeon et al.7 showed that the diurnal variation of plasma levels of 17-hydroxycorticosteroids in night workers and blind persons was the same as that in normal subjects. Hellman et al.8 showed that cortisol was secreted intermittently rather than...
